If using a broad enough definition to include all of Silicon Valley, the answer is clearly San Fransisco.  If not, then D.C.

Cumulatively over US history, the answer has to be Boston, but most of the really important stuff there happened quite early and it had a 100+ year head start on Chicago and the California cities (and even a ~50 year head start on D.C.), so this isn't really fair.

Gonna go with LA.  Massive population.  It has Hollywood, which is what is globally significant in most parts of the world.  There's also a lot of international travel to and from there.  Cities like that get a boost on the global scene even if they aren't quite as significant in the US.  Miami would be another example of a city that is disproportionately significant to the rest of the world (though it's no-where near the second most important US city even by this calculation).

San Francisco is rated as the second-largest global finance center in the United States, only behind New York, so it has my vote.

Second strongest argument is for D.C. though, since so much of the (international) think tank/NGO world is centered there.

Internationally - DC (what American government does influences  (for good or bad) the whole world). Domestically - LA, though Bay Area as a whole could compete.

LA because of how much it interacts with foreign countries (Latin America, Pacific, etc.)

Chicago is up there but it’s mostly an influence on the country and surrounding states then a foreign country
