This is mostly me musing the thought, especially since I know it's a silly question, considering Grassley loves Iowa and Iowa loves him back. But do the conservatives of the state love him?

We all know what's been happening in Iowa recently, and asked whether Grassley would support an amendment, he said "You better ask me in a month, after I've had a chance to think". He goes on to say, "But it doesn't have to be marriage ... There's things like civil unions."

Which is a position governor Culver took right after the ruling last Friday. Say marriage is between a man and a woman, then after the ruling, take a while to think about it and eventually come out in support of it. I don't even think governor Culver came out in support of civil unions before the ruling.

So, I know Grassley hasn't said anything yet, but assuming he doesn't back a marriage amendment, does anything think a farther right Iowa Republican has the balls to challenge the almighty Grassley?

He's not going to be kicked out because he supports civil unions (like many other Republicans). Plus, he's a giant in that state and not some RINO. He'd smoke anyone.

This thread isn't about the fact that he supports civil unions, it's about the possibility of him not supporting a marriage amendment - even if he doesn't back marriage equality. Do you have any notion at all about how the Republican Party of Iowa feels right not about this? They're doing whatever it takes and if Grassley agrees with other statewide officials that they're out of line, then one of them can definitely primary him on this issue alone.

Like Phil said, he's a giant in the state.  Even if he did get a primary challenger, it wouldn't even be a challenge.  Iowa isn't like Virginia, where John Warner faced a serious primary challenge in 1996; there isn't enough of a conservative force in Iowa to actually deny Grassley the nomination.

He could come out as gay and easily survive a primary, so I don't see why his support would seriously affect his nomination chances

Senator Grassley is 76 years old.  In my view, he should not even be running again anyway.

But, to answer your question, no, Grassley will not lose a primary, should anyone run against him in the primary, regardless of Grassley's musings or statements about gay marriage or civil unions.
