If FDR lost or didn't run in 1932, with Hoover winning a second term, what would things be like? Would the Great Depression have ended sooner or later than with FDR's presidency?

The most likely scenario for this happening is Hoover coming back after a long retirement and winning in 1948 (he was still eligible, having only served one term, and would have been grandfathered in anyways, having served before the 22nd Amendment),not winning in 1932.

That second term would have been Hoover doing what he does best in rebuilding Europe, so probably an expanded Marshall Plan.

Al Smith wins in 1932 and fails to do anything to help the Depression, so Hoover manages to make a narrow comeback in 1936, does slightly more to address the Depression than Smith, and retires in 1940.
