Almost certainly.  Occasionally, you'll see a Senator vote Yes on cloture so that there's debate and a final vote, and then vote No on the final bill/nominee.  However, if a Senator doesn't even want to advance the nominee/bill, then they're pretty much always voting No again in the final vote.

CNN's Political Director David Chalian doesn't think Collins, Flake will vote NO tomorrow after okaying the FBI Investigation yesterday.

He also thinks Manchin is a YES given what he says numerous times that he would decide what the people of WV want.

McConnell can and will hold the vote open if needed tomorrow. If the Senate needs to spend 6 or 8 hours voting to wait for Daines, it'll do just that.

I can't see Manchin allowing himself to be the deciding vote in favor of confirmation if Murkowski and Collins both vote no.  So he'll vote the same way as Collins, and those two votes will decide the outcome.
