It depends entirely on how successful Hillary Clinton's presidency is.  If she leaves office with job approval ratings around 50% or better, he might be able to position himself as the rightful heir if he is seen as handling his VP duties well. 

Even under such circumstances however, I doubt Vice President Kaine would scare off major primary opponents.  Sens. Booker, Gillibrand and Klobuchar are all subtly making moves aimed toward the future and if she wins the California Senate race, it is possible that Kamala Harris could join that list as well.  Any number of currently unknown people might emerge over the next eight years.
